I have a total of 6.4 years of exp and I am level 10. I have some medical emergencies but my manager is not agreeing to provide WFH. Instead he asked me to take 6 months LOP. Kindly advise what i need to do now.

Drop him a mail with marking everyone from your TL to Level 6/5 and request for it with valid reasons

I have already sent an email. It's almost 5 days, no reply. If Ping /call also no reply

Try to involve your HRPA, though I understand how baseless they actually work

Since you have an other project team offering to take you in and provide WFH option, considering your manager was fine with you taking an immediate LOP, your manager’s intention is questionable.
However, with no factual information or connection in this matter, my understanding is the following.
Since you are stating medical emergencies, your request for alternative form of work must be considered for review provided you have furnished enough supporting documentation.
Try to loop in the mental health / employee well-being department folks, if you can find their contacts. If that does not work, you may need to loop in your vertical’s leader.

I have searched for a new role. Another project manager agreed to hard lock me. But last night i received an email saying they won't be able to hard lock me, as the current manager is not releasing me

Put resignation and give reason as business leadership. HR and AMD will connect with you. State the same. Also you will have 90 days to search for a job and companies prefer people with less notice period. As far as I know there is no HR policy yet to force you to work from the office until it is mandatory from client.
