Is it mandatory to talk to the supervisor, before dropping resignation mail ?
From where I can resign, Is it from workday portal or directly mail Hr or supervisor?

Don't think it's mandatory but I feel it's a good practice in terms of not surprising them and offending them assuming you don't want to burn bridges before leaving.
I think you can raise a support request to resign, better to ask HR
