Manager can defer only one time. Second time, by default it will be approved.. You need to wait for some time to raise second time. I am not sure about the waiting time between first and second time request.

What if request get auto approved? Will it be closed from backend ?

You profile will be visible in TMP as PE profile. You are eligible to apply for PE eligible SO in TMP. If got selected for PE SO, manager need to release you I think.
